SUMMARY:
We are looking for a Sr. Scientist/Principal Scientist for our Protein Science Team that will be responsible for mammalian expression and purification of therapeutic proteins. The Scientist will have wide-ranging responsibilities and tremendous opportunities within a dynamic, enthusiastic, highly interdisciplinary and collaborative team.
RESPONSIBILITIES:
- Optimize and perform the mammalian expression and purification of engineered fusion protein therapeutics.
- Experience with transient expression from HEK and CHO cells. Stable CHO expression at research scale is preferred.
- Express proteins from and maintain mammalian cell culture at various scales, small-scale high-throughput screening at microgram quantities to milligram quantities.
- Perform protein purification using gravity and AKTA-based methods.
- Purify and characterize expression samples using preparative FPLC and analytical HPLC.
- Perform characterization and quality assessments (e.g. SDS-PAGE, HPLC, endotoxin) of purified proteins.
- Proactively investigate different technologies, methods, and emerging tools to increase work efficiency
- Provide protein science expertise and effectively collaborate with multidisciplinary scientists on cross-functional teams
- Work with CROs, collaborators and partners to transfer samples, data and protocols into, or out of, Xilio, as appropriate.
- Enter all data into laboratory notebooks and databases routinely, generate weekly summary reports for management review, and create sample reports for release of protein lots.
- Regularly present data and analyses at project team and department meetings.
QUALIFICATIONS:
- Ph.D. in Biochemistry or a relevant scientific discipline with 4-7+ years of industry experience or BS/MS with 14 years of experience with therapeutic protein expression and purification.
- Experience in protein expression of multiple molecule types, including antibodies and fusion proteins from mammalian cell lines, including HEK293 and CHO transient and stable expression platforms
- Strong technical expertise with AKTA-based protein purification with various chromatography techniques, including affinity, ion exchange, SEC, HIC, etc.
- Experience with common protein characterization methods, including SDS-PAGE, HPLC, CE-SDS, and endotoxin
- Experience with robotics, high throughput expression and purification platforms and bioanalytical characterization techniques is highly desirable.
- A track record of innovation and scientific rigor, strong problem solving and troubleshooting skills, and ability to contribute constructively to discussions.
- Goal oriented, organized, capable of prioritizing and working on multiple projects, and able to execute to timelines that meet company goals.
- Work well in a cross functional team environment, both independently as well as be willing to help others.
- Good verbal and written communication skills.
