Scribe Therapeutics is a molecular engineering company focused on creating best-in-class in vivo therapies that permanently treat the underlying cause of disease. We are seeking a highly creative, passionate, and motivated individual to join us in our quest to develop the next generation of CRISPR-based therapeutics. The current role is for a Scientist to advance our ongoing efforts to engineer highly active and specific CRISPR enzymes.  The candidate should have a passion for building the CRISPR systems that advance our therapeutic programs. The role will work collaboratively across interdisciplinary teams, including  disease experts and computational biologists. The candidate should bring experience in protein engineering and a passion for creating tools to meet the challenge of this new frontier in genetic medicine.

Key Responsibilities:

  • Design, screen, and characterize engineered CRISPR molecules to improve activity and specificity
  • Develop key new functionalities for our CRISPR-CasX platform
  • Design and execute molecular biology and cloning workflows to generate libraries of protein and RNA variants
  • Design and execute pooled screens to select for molecules with desired attributes
  • Compare and assess protein and RNA variants to produce mechanistic insights to guide engineering efforts
  • Collaborate with NGS and Bioinformatics teams to build robust pipelines 
  • Effectively collaborate with a fully integrated team to facilitate execution on projects within established timelines
  • Foster a driven, fast-paced, dynamic, and fun environment in which to do rigorous science

Required Skills and Background:

  • A PhD in Bioengineering, Biochemistry, Biophysics, Synthetic Biology, Genetics or related discipline.
  • At least 3 years of post-graduate experience working in an academic or industry lab, with a proven track record of expertise in molecular biology, protein engineering, and pooled screening.
  • Experience with protein engineering, including deep mutational scanning, directed evolution, and rational design approaches. 
  • Experience in designing reporter systems, screening in human cells, and readout by FACS and NGS.
  • Experience with assay development for functional characterization of enzymes.
  • Demonstrated quantitative and scientific thinker as evidenced by a strong publication record.
  • Ability to work both independently and collaboratively in a fast-paced, interdisciplinary research team.

Preferred Skills and Background:

  • Experience in engineering CRISPR-Cas systems.
  • Familiarity with CRISPR therapeutic approaches.
  • Experience with functional genomics or next-generation sequencing approaches.
  • Experience with FISH, IF, WB, and other cell-based assays.
  • Working knowledge of statistics to enable the design and analysis of pooled screens with >10K library members.
  • Experience with Rosetta or other packages for computational protein design.

At the time of posting, the base pay wage range for this role is $125,000-170,000 per year.  The offered pay range will depend on internal equity and the candidate’s relevant skills, experience, qualifications, training, and market data.  Additional incentives are provided as part of the complete package in addition to comprehensive medical and other benefits.
