About The Role 

Alltrna is seeking a highly motivated and talented Senior Scientist to join our Cell & Organismal Biology team. The successful candidate will be a key driver in expanding Alltrna’s disease modeling platform, and use state-of-the-art technologies to engineer, differentiate and utilize clonal CRISPR/Cas9-edited hPSC lines to enhance the throughput and physiological relevance of disease models. This role provides a unique opportunity to apply human pluripotent stem cell research to support drug discovery and therapeutic development.  The candidate will have a proven track-record of scientific excellence as evidenced by a strong publication and/or patent record. She/he will work with multifunctional groups to make research strategies, design, and perform experiments to evaluate new technologies while supervising and training junior team members. He/she should be a strong team player, clear communicator, enjoy scientific discovery and work effectively in a collaborative interdisciplinary environment.

Core Responsibilities:

  • Establish efficient and robust protocols to generate various specific cell types (hepatocytes, neuron, RPE, or muscle) from human pluripotent cells to model human diseases
  • Develop disease phenotype-specific molecular and functional assays such as enzymatic activity, receptor function, metabolic assays in relevant cell types differentiated from human pluripotent stem cells
  • Introduce disease-causing mutations using CRISPR/Cas9 technology or other related gene-editing platforms
  • Design and execute experiments, interpret experimental data, establish workflows including standard cell biology and stem cell biology methods
  • Design and execute preclinical in vitro and ex vivo experiment to support both mechanism of action studies and the clinical translation for engineered tRNA therapeutic development
  • Continually cultivate scientific/technical expertise through critical review of the scientific literature, attending external conferences, and developing relationships with key opinion leaders

Qualifications and Skills:

  • PhD in cell & molecular biology, chemical biology, or a related discipline with 6+ years of experience (for Sr. Scientist) in biotech or pharmaceutical company
  • Proven expertise in developing novel human stem cell approaches, applications to unique biological problems, profiling, and assay development
  • Deep knowledge of human pluripotent stem cell culture, characterization, QC, and differentiation (hepatocytes, neuron, RPE, or muscle)
  • Strong background and hands-on experience in hepatic biology
  • Proven hands-on experience with a variety cell-based and molecular biology techniques, such as FACS, flow cytometry, RT-qPCR, ELISA, immunostaining, high-content and confocal imaging, genetic and epigenetic assessment
  • Extensive experience with gene editing applications, e.g., gene knock-out, targeted knock-in, multiplexed gene editing, and base editing)
  • Experience in technique for genetic manipulation using siRNA, lentivirus, or PiggyBac system
  • Demonstrated ability to work in a dynamic environment as a team player with a strong work ethic, excellent communication, and time management skills
  • Foster a collaborative, creative, and rigorous culture of scientific discovery

Preferred Skills:

  • Experience with microphysiological systems including 3D multi-cellular culture models, organoids, bioprinting, organ-on-a-chip models
  • Experience designing AAV expression vectors, gene delivery in vitro and in vivo is a plus.
